JB-4 Plus® Embedding Kit is a HEMA-based glycol methacrylate (GMA) embedding system for preparation of semi-thin plastic sections for light microscopy. The formulation is designed for histological workflows that benefit from controlled, low-temperature polymerization. Cured blocks can be sectioned at approximately 0.5–3.0 µm or thicker for routine histology, special staining, and histochemical procedures.

Polymerization is exothermic. Cooling on ice or at approximately 4°C slows the reaction and limits heat generation during curing; under these conditions, polymerization may require several hours to overnight.

JB-4 Plus® processing does not require xylene or chloroform clearing. Many soft-tissue specimens also do not require dehydration to absolute alcohol, although dense, bloody, or fatty specimens may require more complete dehydration.

Applications

Semi-Thin Histology

JB-4 Plus® is used to prepare plastic-embedded specimens for semi-thin light-microscopy sectioning. Sections approximately 0.5–3.0 µm or thicker can be prepared using a microtome and knife suitable for plastic-embedded material.

Routine & Special Staining

JB-4 Plus® sections can be used with routine histological stains and many special staining procedures. Because the polymerized glycol methacrylate remains within the section, staining conditions may require adjustment for section thickness and stain chemistry.

Histochemistry

Many histochemical procedures can be performed directly on JB-4 Plus® sections. The retained polymer should be considered when selecting reagents with relatively high molecular weight or limited penetration through the polymer matrix.

Temperature-Controlled Embedding

Polymerization on ice or at approximately 4°C slows curing and reduces the rate of heat generation. This approach is useful when control of the polymerization exotherm is important to specimen preparation.

Fixation, Dehydration & Infiltration

Specimens can be fixed with 10% neutral buffered formalin or another routine histological fixative. For workflows requiring a methanol-free formaldehyde-based fixative for light and electron microscopy, see Poly/LEM Fixative.

  • Fixation can be performed at room temperature or 4°C
  • Dehydration can be performed at room temperature or 4°C
  • Absolute alcohol is not required for many soft-tissue specimens
  • Dense, bloody, or fatty tissues may require more complete dehydration
  • Do not introduce JB-4 Plus® resin into a routine histology tissue processor unless the instrument manufacturer specifically permits polymer infiltration
  • Avoid unnecessary heat and direct light during infiltration

To prepare 100 mL of infiltration solution, combine 100 mL of Solution A with 1.25 g of plasticized benzoyl peroxide catalyst. Accurate catalyst measurement is important because catalyst concentration affects polymerization rate and the associated exotherm.

The catalyzed infiltration solution can be stored for up to two weeks in a cool, dark location or under refrigeration at 4°C.

Embedding & Polymerization

  • Prepare fresh catalyzed infiltration solution for the embedding mixture
  • Do not use previously used infiltration solution to prepare the embedding mixture
  • For 25 mL of freshly prepared infiltration solution, add exactly 1.0 mL of Solution B
  • Mix thoroughly and begin embedding immediately after Solution B is added
  • Polymerize under oxygen-excluded conditions
  • Use block holders, an airtight covering, or an appropriate vacuum method to minimize oxygen exposure during polymerization
  • Use cooling at approximately 4°C when slower polymerization and lower heat generation are required

Do not pre-cool embedding molds before filling, because condensation can interfere with uniform polymerization at the block surface.

JB-4® Plastic Block Holders can be used to limit oxygen exposure at the block face during polymerization. Compatible Embedding Mold Trays are available for JB-4® and JB-4 Plus® workflows.

Sectioning & Staining

  • Use a microtome designed for plastic-embedded specimens
  • Glass, Ralph-type, or tungsten carbide knives can be selected according to block dimensions and required section thickness
  • Sections can be prepared at approximately 0.5–3.0 µm or thicker
  • Polymerized glycol methacrylate cannot be completely removed from the section
  • Routine histological stains and most histochemical procedures can be performed on the retained polymer section
  • High-molecular-weight staining reagents and immunohistochemical reagents may show incomplete penetration through the polymer matrix

Immunohistochemistry & Bone Considerations

JB-4 Plus® is not recommended for routine immunohistochemistry because the polymerized glycol methacrylate cannot be completely removed from the section and may restrict antibody access to antigen sites.

JB-4 Plus® can be used with bone specimens; however, the cured polymer is slightly softer than standard JB-4® Embedding Kit and Osteo-Bed formulations. For most bone procedures requiring a harder embedding matrix, standard JB-4® or a dedicated mineralized-tissue system is preferred. Application Tips

  • Measure catalyst and accelerator accurately to maintain reproducible polymerization kinetics
  • Keep infiltration and embedding solutions away from unnecessary heat and direct light
  • Prepare fresh embedding solution immediately before use
  • Exclude oxygen during polymerization to support complete curing at the block surface
  • Use cooling when control of polymerization rate and exotherm is important
  • Keep fixation, dehydration, infiltration, and curing conditions consistent when comparing specimens

Handling & Storage

  • Store the complete kit at 4°C according to current kit labeling
  • When components are stored separately, follow the applicable component SDS
  • Solution A: store at 4°C in a cool, well-ventilated location
  • Solution B: store at room temperature with the container tightly closed in a cool, well-ventilated location
  • Plasticized benzoyl peroxide catalyst: store at 5 ± 5°C, protected from sunlight and segregated from combustible materials
  • Use all components with appropriate laboratory ventilation
  • Wear protective gloves and eye protection during handling
