Luxol Fast Blue, Ready-to-Use

Luxol Fast Blue (LFB), Ready-to-Use is a histological staining solution for demonstrating myelin in central nervous system (CNS) tissue. After staining and differentiation, myelinated white matter appears blue, providing contrast with gray matter and regions containing reduced or absent myelin.

This ready-to-use Luxol Fast Blue solution is supplied in a 500 mL bottle and stains up to approximately 200 slides. The method is commonly used with formalin-fixed, paraffin-embedded CNS tissue.

Applications

Myelin Visualization

Luxol Fast Blue is used to visualize myelin in central nervous system tissue. Following appropriate differentiation, myelinated white matter stains blue and is readily distinguished from gray matter.

Visualization of Myelin Loss

Regions with reduced or absent myelin show correspondingly reduced or absent Luxol Fast Blue staining, providing histological contrast with surrounding myelinated tissue.

Staining & Preparation Tips

• Use adequately fixed paraffin sections; fixation for at least 24 hours in 10% neutral buffered formalin is recommended
• Deparaffinize sections before staining
• Incubate slides in Luxol Fast Blue at 56°C overnight
• Keep the staining container tightly capped during overnight incubation to limit evaporation of the alcoholic staining solution
• Differentiate with Scott's Bluing Reagent or prepared lithium carbonate solution for 10-20 seconds, followed by 70% ethanol until gray matter can be distinguished from blue-stained white matter
• Check differentiation microscopically and repeat the differentiation steps if necessary
• Do not allow tissue sections to dry during differentiation
• After dehydration and clearing, coverslip with a synthetic resin mounting medium such as Poly-Mount® Xylene

• Use in a well-ventilated laboratory area
