The Methyl Methacrylate-Butyl Methacrylate Embedding Kit (MMA/BMA Kit), Cat. 03573, is an acrylic resin system for embedding tissue for thin or thick sectioning. Blends of methyl methacrylate (MMA) and n-butyl methacrylate (BMA) produce clear, firm embedding blocks whose hardness can be adjusted by changing the MMA:BMA ratio.

Increasing the proportion of BMA produces a softer block. The resin may be polymerized thermally with plasticized benzoyl peroxide or photopolymerized with benzoin methyl ether and long-wave ultraviolet light.

For electron microscopy applications, the MMA/BMA resin may be used with an added crosslinking agent such as divinylbenzene (DVB). DVB is not included in Cat. 03573 and should be added only according to an appropriate validated embedding procedure.

Applications

Tissue Embedding for Sectioning

The MMA/BMA system is used to embed fixed and dehydrated tissue for subsequent thin or thick sectioning. The relative amounts of MMA and BMA can be adjusted to modify block hardness for the intended specimen and sectioning procedure.

Electron Microscopy Embedding

MMA/BMA embedding media may be adapted for electron microscopy by adding an appropriate crosslinking agent such as divinylbenzene. The crosslinker is not supplied with the kit, and concentration and processing conditions should follow the selected EM protocol.

Thermal Polymerization

For thermal curing, Technical Data Sheet #408 specifies MMA:BMA ratios of 8:2 or 9:1 with 1% w/v benzoyl peroxide. Polymerization is performed overnight at approximately 60°C.

UV Polymerization

For photopolymerization, the same MMA:BMA ratios may be used with 0.5% benzoin methyl ether. Long-wave UV illumination around 360 nm is used, with exposure time dependent on lamp output, distance from the specimen, block geometry, and temperature.

Compatibility & Bonding Considerations

Polymerization rate, exotherm, block hardness, and sectioning behavior depend on monomer ratio, initiator concentration, resin volume, temperature, and curing conditions. Larger quantities of catalyzed resin can polymerize more rapidly and generate greater heat.

Application Tips

  • For thermal curing, use the specified MMA:BMA ratio with 1% w/v plasticized benzoyl peroxide and polymerize overnight at approximately 60°C
  • For UV curing, use 0.5% benzoin methyl ether with an MMA:BMA ratio of 8:2 or 9:1 and long-wave UV illumination around 360 nm
  • Position molds so UV light can penetrate the complete block during photopolymerization
  • Use closed BEEM capsules, gelatin capsules, or other closed embedding containers as specified in the procedure
  • Keep final cast volumes as small as practical because larger quantities can polymerize more rapidly and generate temperatures capable of damaging tissue structure
  • Colder photopolymerization conditions require longer curing times
